产品介绍This gene encodes a cell adhesion protein which is a member of the immunoglobulin superfamily. The encoded protein is involved in cell-to-cell interactions as well as cell-matrix interactions during development and differentiation. The encoded protein has been shown to be involved in development of the nervous system, and for cells involved in the expansion of T cells and dendritic cells which play an important role in immune surveillance. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2011]

靶点与功能

NCAM1（Neural Cell Adhesion Molecule 1，CD56）是免疫球蛋白超家族成员，通过糖基化修饰（含多唾液酸链）介导细胞间粘附与信号传递，核心功能包括：

神经发育：调控神经元迁移、轴突生长及突触形成（多唾液酸-NCAM1缺失导致海马突触密度降低30%）；

肿瘤标志物：在神经母细胞瘤（阳性率95%）、小细胞肺癌（70%）、NK/T细胞淋巴瘤中特异性高表达，是临床病理诊断的重要指标；

免疫调节：NK细胞表面CD56分子参与细胞毒性激活（CD56bright亚群分泌细胞因子，CD56dim亚群介导靶细胞杀伤）。

实验操作关键要点

FCM检测NK细胞（外周血样本）：

样本处理：全血经红细胞裂解液（BD FACS Lysing Solution）处理后，PBS洗涤2次，调整细胞浓度至1×10⁶/mL；

抗体孵育：CD56-PE标记抗体（BD 555516）1:20稀释，4℃避光孵育30分钟，流式细胞仪检测CD56⁺CD3⁻ NK细胞比例（健康人外周血占比7-15%）。

IHC-P神经母细胞瘤染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），肿瘤细胞胞膜信号显著；

阳性定位：神经母细胞瘤细胞胞膜/胞质呈棕黄色阳性，与Synaptophysin共染可提高诊断特异性（双阳性率>90%）；

对照设置：正常肾上腺组织（阴性对照）和神经节细胞瘤（阳性对照）。

WB验证（脑皮层组织）：

样本制备：小鼠脑皮层用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：8% SDS-PAGE凝胶（因亚型分子量差异大），恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（300mA恒流2.5小时）；

抗体孵育：一抗1:500稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光时间60秒（可见120/140/180kDa多条带）。
