产品介绍HIV-1, the causative agent of acquired immunodeficiency syndrome (AIDS), contains an RNA genome that produces a chromosomally integrated DNA during the replicative cycle. Activation of HIV-1 gene expression by the transactivator Tat is dependent on an RNA regulatory element (TAR) located downstream of the transcription initiation site. The protein encoded by this gene is a transcriptional repressor that binds to chromosomally integrated TAR DNA and represses HIV-1 transcription. In addition, this protein regulates alternate splicing of the CFTR gene. A similar pseudogene is present on chromosome 20. [provided by RefSeq, Jul 2008]

靶点与功能

微管相关蛋白（MAPs）是一类通过结合微管调控其稳定性和动态性的关键蛋白，主要包括 Tau、MAP2、MAP4 等家族成员：

Tau蛋白：通过磷酸化状态调控微管稳定性，过度磷酸化会导致神经纤维缠结，与阿尔茨海默病等神经退行性疾病密切相关；

MAP2：主要表达于神经元树突，参与树突形态维持和微管网络组织；

MAP4：广泛存在于非神经细胞，在细胞分裂（纺锤体形成）和应激反应中起核心作用。

这些蛋白的异常表达或修饰与肿瘤细胞侵袭、细胞周期紊乱及神经病变直接相关。

实验操作关键要点

WB优化（以Tau磷酸化检测为例）：

样本处理：脑组织或神经细胞需用含 磷酸酶抑制剂（如Na3VO4） 的RIPA裂解液冰上裂解40分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保磷酸化Tau（~60kDa）与非磷酸化条带分离；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（神经退行性疾病研究）：

抗原修复：采用 EDTA缓冲液（pH8.0） 高压修复5分钟（针对福尔马林固定石蜡切片），增强磷酸化Tau信号；

阳性对照：推荐使用阿尔茨海默病患者海马组织切片（神经元内可见棕黄色神经纤维缠结）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。
