产品介绍Tau proteins are important Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization. Tau proteins subcellular located in the axons of neurons, in the cytoso l and in association with plasma membrane components. It expressed in neurons. PNS-tau is expressed in the peripheral nervous system while the others are expressed in the central nervous system.

MAPT（微管相关蛋白Tau）基因编码的Tau蛋白，主要功能是稳定神经元微管结构、促进轴突运输，通过可变剪接产生6种亚型（3R/4R，含3-4个微管结合重复序列）。在病理条件下，Tau蛋白发生过度磷酸化（如Ser396/Thr231位点），从微管解离并聚集形成神经纤维缠结（NFTs），这是阿尔茨海默病（AD）、额颞叶痴呆（FTD）等“Tau蛋白病”的核心病理特征。

实验操作关键要点

WB检测Tau磷酸化状态：

样本处理：脑组织需用含磷酸酶抑制剂（如PhosSTOP）的RIPA裂解液冰上裂解40分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，可同时检测总Tau（如Tau-5抗体）和磷酸化Tau（如pS396抗体）；

抗体孵育：一抗用5% BSA+TBST按1:1000稀释，4℃摇床孵育过夜，HRP标记二抗（1:5000）室温孵育1小时。

IHC-P检测神经纤维缠结：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），自然冷却至室温；

阳性对照：推荐使用AD患者海马组织切片（神经元胞质内可见棕黄色NFTs）；

封闭处理：3% H₂O₂室温孵育10分钟阻断内源性过氧化物酶，5% BSA封闭30分钟。

研究与临床应用

疾病机制研究：通过双标IF（如Tau+β-淀粉样蛋白）揭示AD病理级联反应，或联合GSK-3β/CDK5激酶抑制剂验证Tau磷酸化调控通路；

药物开发：靶向病理性Tau的单克隆抗体（如Eisai的Etalanetug、Merck的MK-2214）通过促进聚集体清除或抑制种子传播，已进入AD临床试验；

诊断工具：开发的Tau抗体试剂盒，可检测脑脊液/血液中pg级Tau蛋白，线性范围7.8-500pg/mL，助力AD早期诊断。
