产品介绍Tau proteins are important Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization. Tau proteins subcellular located in the axons of neurons, in the cytoso l and in association with plasma membrane components. It expressed in neurons. PNS-tau is expressed in the peripheral nervous system while the others are expressed in the central nervous system.

靶点与功能

TFF3（Trefoil Factor 3）是小分子分泌型糖蛋白（含特征性“三叶草”结构域），核心功能与研究价值包括：

黏膜保护与修复：由肠黏膜杯状细胞分泌，通过与黏液糖蛋白结合增强黏液凝胶稳定性，在炎症性肠病（IBD）、胃溃疡中促进上皮细胞迁移和创面愈合（动物实验显示TFF3过表达可使溃疡愈合速度提升2倍）；

肿瘤标志物：在结直肠癌（阳性率78%）、胃癌、胆管癌中高表达，与肿瘤分化程度负相关（低分化腺癌表达量是高分化的3.5倍），可通过IHC检测辅助病理诊断；

激素调节作用：在乳腺组织中受雌激素调控，与三阴性乳腺癌的内分泌治疗耐药相关，可作为潜在治疗靶点。

实验操作关键要点

IHC-P结直肠癌组织染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），肿瘤细胞胞质呈棕黄色颗粒状阳性（黏液腺癌表达最强）；

阳性对照：正常结肠黏膜（杯状细胞强阳性）；

注意事项：避免使用EDTA修复液（螯合Ca²⁺影响三叶草结构域稳定性）。

WB检测（肠黏膜样本）：

样本处理：大鼠小肠黏膜用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，14000g离心20分钟取上清；

电泳条件：15% SDS-PAGE凝胶（Tris-Tricine缓冲系统），恒压80V浓缩胶、120V分离胶，电泳60分钟（小分子蛋白分离优化）；

转膜技巧：使用0.22μm PVDF膜，转膜缓冲液添加20%甲醇，恒流300mA转膜45分钟。

ELISA双抗夹心法（血清样本）：

包被抗体（重组单抗，2μg/mL）4℃包被过夜，血清样本1:50稀释，检测抗体（生物素标记多抗，0.5μg/mL）37℃孵育1小时，标准曲线范围0.1-100 ng/mL（检测下限可达0.05 ng/mL）。
