产品介绍Members of the trefoil family are characterized by having at least one copy of the trefoil motif, a 40-amino acid domain that contains three conserved disulfides. They are stable secretory proteins expressed in gastrointestinal mucosa. Their functions are not defined, but they may protect the mucosa from insults, stabilize the mucus layer and affect healing of the epithelium. This gene is expressed in goblet cells of the intestines and colon. This gene and two other related trefoil family member genes are found in a cluster on chromosome 21. CD71（即转铁蛋白受体1，TFRC）是一种II型跨膜糖蛋白，通过介导转铁蛋白-铁复合物的内吞作用调控细胞铁摄取，维持铁稳态。其功能与细胞增殖高度相关：在造血干细胞、免疫细胞等快速分裂细胞中高表达，更在肺癌、结直肠癌、胰腺癌等肿瘤细胞中异常过表达（较正常细胞高3-10倍），成为肿瘤增殖活性和预后评估的重要标志物。

实验操作关键要点

WB优化：

样本处理：肿瘤细胞裂解液需添加10mM DTT（破坏二硫键，避免多聚体干扰），冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保95kDa主条带清晰；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强胞膜信号；

阳性对照：建议使用结肠癌组织切片（肿瘤细胞胞膜呈棕黄色阳性）；

封闭处理：5%驴血清室温封闭30分钟，减少非特异性结合。

FC检测要点：

细胞制备：单细胞悬液经0.2% BSA-PBS洗涤2次，避免使用EDTA（影响受体构象）；

抗体孵育：1μg抗体/1×10⁶细胞，4℃避光孵育30分钟，流式缓冲液洗涤后上机检测。
