产品介绍Oct2 is a transcription factor that specifically binds to the octamer motif (5'-ATTTGCAT-3'). Oct2 regulates transcription in a number of tissues in addition to activating immunoglobulin gene expression. It also modulates transcription transactivation by NR3C1, AR and PGR. Oct2 is B cell specific, belongs to the POU transcription factor family class 2 subfamily and contains 1 homeobox domain.

靶点与功能

P-糖蛋白（P-glycoprotein，P-gp，ABCB1/MDR1）是定位于细胞膜的ATP依赖型外排泵，核心功能与研究价值包括：

多药耐药（MDR）机制：通过ATP水解能量将化疗药物（如阿霉素、紫杉醇、长春新碱） 泵出细胞，降低胞内药物浓度，导致肿瘤对多种结构无关药物产生耐药性（IC50值升高5-50倍）；

生理屏障作用：在血脑屏障、肠道上皮、肝胆管等高表达，参与外源性物质清除（如肠道药物吸收减少50%以上）；

肿瘤预后意义：乳腺癌、肺癌、卵巢癌中P-gp过表达患者化疗响应率降低40%，无进展生存期缩短35%（Clin Cancer Res, 2024）。

实验操作关键要点

WB检测（耐药细胞系）：

样本处理：A2780/ADR（阿霉素耐药卵巢癌细胞）用含1mM Na3VO4（磷酸酶抑制剂） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：7.5% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（300mA恒流2小时，冰浴）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，耐药株条带强度显著高于亲本株（A2780）。

IHC-P肿瘤组织染色：

抗原修复：EDTA缓冲液（pH9.0）高压修复15分钟（石蜡切片），细胞膜信号增强；

阳性定位：耐药肿瘤组织细胞膜呈棕黄色线性阳性（如肺腺癌耐药灶），正常组织中肝胆管上皮细胞阳性可作为内参；

对照设置：敲除小鼠组织（阴性对照）和Caco-2细胞（人结直肠腺癌细胞，高表达P-gp，阳性对照）。

IF双标实验（血脑屏障模型）：

与ZO-1抗体共染（紧密连接标志物），使用Alexa Fluor 488（绿色，P-gp膜定位）和594（红色，ZO-1），激光共聚焦观察脑微血管内皮细胞顶膜共定位（药物外排功能结构基础）。
