产品介绍This gene encodes a member of the p53 family of transcription factors. An animal model, p63 -/- mice, has been useful in defining the role this protein plays in the development and maintenance of stratified epithelial tissues. p63 -/- mice have several developmental defects which include the lack of limbs and other tissues, such as teeth and mammary glands, which develop as a result of interactions between mesenchyme and epithelium. Mutations in this gene are associated with ectodermal dysplasia, and cleft lip/palate syndrome 3 (EEC3); split-hand/foot malformation 4 (SHFM4); ankyloblepharon-ectodermal defects-cleft lip/palate; ADULT syndrome (acro-dermato-ungual-lacrimal-tooth); limb-mammary syndrome; Rap-Hodgkin syndrome (RHS); and orofacial cleft 8. Both alternative splicing and the use of alternative promoters results in multiple transcript variants encoding different proteins. Many transcripts encoding different proteins have been reported but the biological validity and the full-length nature of these variants have not been determined. [provided by RefSeq, Jul 2008].

靶点与功能

Pan-Actin识别的肌动蛋白（Actin）是真核细胞中含量最丰富的蛋白质（占细胞总蛋白5%-10%），核心特性包括：

结构保守性：α-肌动蛋白（肌肉）、β/γ-肌动蛋白（非肌肉）的氨基酸序列同源性＞90%，泛抗体通过识别保守C端表位实现广谱结合；

内参金标准：在多数组织/细胞中表达稳定（CV＜5%），分子量42kDa位于WB常用区间，与多数目的蛋白无重叠；

细胞功能标志：参与细胞迁移（应力纤维）、胞质分裂（收缩环）、信号转导（Rho-GTPase通路），荧光标记可直观显示细胞骨架动态。

实验操作关键要点

WB内参标准化（通用流程）：

上样量控制：推荐20-30μg总蛋白/泳道，5%浓缩胶+10%分离胶（恒压80V→120V），转膜250mA恒流1小时（0.45μm PVDF膜）；

抗体稀释：一抗1:2000稀释（5%脱脂奶粉+TBST），室温孵育1小时（无需4℃过夜），曝光时间10-30秒（42kDa处条带清晰无拖尾）；

数据校正：目的蛋白灰度值需除以Actin灰度值，消除上样误差（如计算“目的蛋白/Actin比值”进行组间比较）。

IF细胞骨架染色：

固定与透化：4%多聚甲醛室温固定15分钟，0.1% Triton X-100透化5分钟（保持微丝结构完整）；

荧光标记：一抗1:500稀释（1% BSA+PBS），4℃孵育过夜，Alexa Fluor 488标记二抗（绿色），DAPI复染细胞核（蓝色）；

典型结构：贴壁细胞可见应力纤维（长纤维状）、皮层肌动蛋白（细胞边缘），迁移细胞前缘可见丝状伪足。

IHC-P组织切片染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），所有细胞胞质呈阳性；

阳性对照：任何组织切片均应呈阳性（肌动蛋白普遍表达），阴性对照需用Actin敲除细胞系验证；

注意事项：避免使用肌肉组织（α-actin高表达可能掩盖其他亚型信号）。
