产品介绍This gene encodes a membrane-bound glycoprotein. The encoded protein acts as an enzyme and plays a central role in thyroid gland function. The protein functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Multiple transcript variants encoding distinct isoforms have been identified for this gene, but the full-length nature of some variants has not been determined. [provided by RefSeq, May 2011].

靶点与功能

TPO（Thyroid Peroxidase）是甲状腺滤泡上皮细胞顶膜的含铁血红素酶（分子量105kDa），通过催化酪氨酸碘化及偶联反应合成甲状腺激素（T3、T4）。其核心临床意义包括：

疾病诊断：TPO抗体是桥本甲状腺炎（HT）的**指标，阳性率＞90%，滴度与甲状腺破坏程度正相关；

风险预测：健康人群中TPO抗体阳性者，未来5年内发生甲减的风险是阴性者的20倍；

研究价值：用于自身免疫性甲状腺疾病动物模型的造模验证，或评估免疫调节药物的疗效。

实验操作关键要点

ELISA定量检测（临床血清样本）：

包被条件：兔单抗用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被16小时，PBST洗涤3次；

样本处理：血清1:100稀释（含1% BSA的PBS），37℃孵育2小时（避免高脂样本干扰）；

显色控制：HRP标记二抗（山羊抗人IgG）1:8000稀释，TMB显色15分钟，2M H₂SO₄终止，450nm读取OD值（参考范围：＜60 IU/mL为阴性）。

IHC-P染色（甲状腺组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），自然冷却至室温；

阳性定位：甲状腺滤泡上皮细胞胞膜/胞质呈棕黄色颗粒状阳性，桥本甲状腺炎可见弥漫性强阳性；

对照设置：必须包含正常甲状腺组织（弱阳性）和非甲状腺组织（如肝脏）（阴性对照）。

WB验证（甲状腺细胞裂解液）：

样本制备：FRTL-5甲状腺细胞用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（250mA恒流90分钟）；

抗体孵育：小鼠单抗1:800稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时。
