产品介绍This gene belongs to the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function. Transcription of this gene is highly inducible in response to many cytokines and hormones. In addition, the expression from some but not all inactive X chromosomes suggests that this gene inactivation is polymorphic in human females. This gene is located within intron 6 of the synapsin I gene and is transcribed in the opposite direction. [provided by RefSeq].

靶点与功能

DNA拓扑异构酶Ⅱ（Topoisomerase II, TOP2）是一类催化DNA双链断裂与重连的核酶，通过改变DNA超螺旋状态参与DNA复制、转录、染色体分离等关键过程。人类TOP2分为α（TOP2A）和β（TOP2B）两种亚型，其中TOP2A在增殖细胞中高表达，是蒽环类、依托泊苷等化疗药物的主要靶点，其表达水平与肿瘤恶性程度及药物敏感性密切相关。

实验操作关键要点

WB优化：

样本处理：肿瘤细胞需用含PMSF和磷酸酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、100V分离胶，确保174kDa条带与Marker分离；

抗体孵育：一抗用5% BSA+TBST按1:1000稀释，4℃摇床孵育过夜，HRP标记二抗（1:5000）室温孵育1小时。

IHC-P注意事项：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），自然冷却至室温；

阳性对照：推荐使用结肠癌细胞系（如HCT116）或胃癌组织切片（细胞核呈强阳性）；

显色控制：DAB显色3-5分钟，避免背景过深（因TOP2A在增殖活跃细胞中高表达）。
