产品介绍This gene encodes a protein initially identified as a thyroid-specific transcription factor. The encoded protein binds to the thyroglobulin promoter and regulates the expression of thyroid-specific genes but has also been shown to regulate the expression of genes involved in morphogenesis. Mutations and deletions in this gene are associated with benign hereditary chorea, choreoathetosis, congenital hypothyroidism, and neonatal respiratory distress, and may be associated with thyroid cancer. Multiple transcript variants encoding different isoforms have been found for this gene. This gene shares the symbol/alias 'TFF1' with another gene, transcription termination factor 1, which plays a role in ribosomal gene transcription. [provided by RefSeq, Apr 2011]

靶点与功能

TTF-1（NKX2-1）是NKX2同源域转录因子家族成员，分子量约38kDa，通过结合甲状腺特异性基因（如甲状腺球蛋白、甲状腺过氧化物酶）和肺表面活性蛋白基因的启动子，调控甲状腺滤泡细胞分化、肺发育及表面活性物质稳态。其核心功能包括：

组织特异性标志物：在甲状腺滤泡细胞、肺Ⅱ型肺泡上皮细胞及Clara细胞中高表达，是甲状腺与肺肿瘤的“身份标签”；

临床诊断价值：肺腺癌（约70%-80%阳性）与鳞癌（约5%阳性）的鉴别指标，甲状腺乳头状癌/滤泡癌（阳性）与髓样癌（阴性）的分型依据；

研究意义：参与肺癌细胞上皮-间质转化（EMT）调控，其缺失与肿瘤侵袭转移及不良预后相关。

实验操作关键要点

IHC-P肺癌分型检测（石蜡切片）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复3分钟（针对福尔马林固定样本），增强核信号；

阳性判断：肺腺癌中腺上皮细胞核呈棕黄色阳性，鳞癌多为阴性（需联合p40/p63检测）；

对照设置：必须包含正常甲状腺组织（强阳性对照）和肺鳞癌组织（阴性对照）。

WB验证（细胞裂解液）：

样本处理：NCI-H1299细胞（肺腺癌，TTF-1阳性）用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：鼠单抗1:500稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光45秒（目标条带约38kDa）。
