产品介绍This gene encodes a member of the paired box (PAX) family of transcription factors. The central feature of this gene family is a novel, highly conserved DNA-binding motif, known as the paired box. Paired box transcription factors are important regulators in early development, and alterations in the expression of their genes are thought to contribute to neoplastic transformation. This gene encodes the B-cell lineage specific activator protein that is expressed at early, but not late stages of B-cell differentiation. Its expression has also been detected in developing CNS and testis and so the encoded protein may also play a role in neural development and spermatogenesis. This gene is located at 9p13, which is involved in t(9;14)(p13;q32) translocations recurring in small lymphocytic lymphomas of the plasmacytoid subtype, and in derived large-cell lymphomas. This translocation brings the potent E-mu enhancer of the IgH gene into close proximity of the PAX5 promoter, suggesting that the deregulation of transcription of this gene contributes to the pathogenesis of these lymphomas. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2013]

靶点与功能

PAX8（Paired Box Gene 8）是PAX家族转录因子，含高度保守的配对盒DNA结合域（128个氨基酸），通过以下机制发挥作用：

胚胎发育调控：在甲状腺、肾脏、 Müller管（输卵管、子宫）的胚胎形成中起“器官规格因子”作用，敲除PAX8导致甲状腺缺如和肾脏发育不全（Nature, 2023）；

肿瘤标志物价值：在甲状腺癌（95%）、卵巢癌（85%）、肾细胞癌（72%） 中特异性高表达，是病理诊断中鉴别甲状腺转移癌与肺腺癌的金标准（阳性率差异＞90%）；

分子机制：通过激活NIS（钠碘同向转运体）、E-cadherin等靶基因，维持肿瘤细胞的分化表型和侵袭能力。

实验操作关键要点

IHC-P病理诊断（甲状腺癌样本）：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），甲状腺滤泡上皮细胞核信号显著增强；

阳性定位：正常甲状腺滤泡上皮细胞核呈棕黄色强阳性，甲状腺乳头状癌中核染色弥漫且强度增加（H-score＞200）；

鉴别诊断价值：在肺转移性腺癌中PAX8阴性（与TTF-1联合检测可提高准确率至98%）。

WB验证（肾癌细胞系）：

样本制备：ACHN细胞（高表达PAX8）用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（250mA恒流1小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，曝光时间45秒（45kDa处可见清晰核蛋白条带）。

IF核定位实验：

细胞处理：HEK293T细胞转染PAX8过表达质粒后，4%多聚甲醛固定15分钟，0.3% Triton X-100透化10分钟；

共定位标记：与DAPI（细胞核染料） 共染，激光共聚焦观察PAX8在核内的点状分布（转录激活区域）。
