It is the rate-limiting enzyme in the synthesis of catecholamines, hence plays a key role in the physiology of adrenergic neurons. Mutations in this gene have been associated with autosomal recessive Segawa syndrome. Alternatively spliced transcript variants encoding different isoforms have been noted for this gene. VEGF（Vascular Endothelial Growth Factor）是一种高度特异性的血管内皮细胞促分裂因子，通过与血管内皮生长因子受体（VEGFR）结合，调控血管通透性增加、内皮细胞增殖迁移及新生血管形成。在肺癌、结直肠癌、乳腺癌等实体瘤中，VEGF过表达会促进肿瘤血管生成，为肿瘤生长提供营养，是抗肿瘤治疗的重要靶点。

实验操作关键要点

WB优化：

样本处理：肿瘤组织需用含蛋白酶抑制剂（如PMSF）的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保45kDa二聚体条带清晰；

抗体孵育：一抗用5% BSA+TBST按1:1000稀释，4℃孵育过夜，HRP标记二抗（1:5000）室温孵育1小时。

IHC-P注意事项：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波加热至95℃，持续20分钟后自然冷却，增强胞质/胞膜信号；

封闭步骤：5%驴血清（PBS稀释）室温封闭30分钟，减少非特异性结合；

阳性对照：建议使用结肠癌组织切片（肿瘤间质血管内皮细胞呈阳性）。
