产品介绍Vascular endothelial growth factor (VEGF), originally known as vascular permeability factor (VPF), is a signal protein produced by cells that stimulates the formation of blood vessels. To be specific, VEGF is a sub-family of growth factors, the platelet-derived growth factor family of cystine-knot growth factors. They are important signaling proteins involved in both vasculogenesis (the de novo formation of the embryonic circulatory system) and angiogenesis (the growth of blood vessels from pre-existing vasculature).

靶点与功能

Villin是一种肌动蛋白结合蛋白，主要分布于肠上皮刷状缘、肾近曲小管、胆囊上皮等高度极性细胞，通过调控微丝骨架动态参与细胞形态维持和物质转运。在结直肠癌、胃癌、胆管癌等上皮源性肿瘤中表达异常，其单克隆抗体可特异性识别肿瘤组织中的Villin蛋白，为肿瘤组织来源鉴定提供可靠依据。

实验操作关键要点

IHC-P优化：

抗原修复：推荐EDTA缓冲液（pH9.0）高压修复3分钟（针对石蜡切片），可显著增强胞膜/胞质信号；

封闭步骤：5%驴血清（PBS稀释）室温封闭30分钟，减少非特异性结合；

显色控制：DAB显色5-8分钟，镜下观察至刷状缘区域呈棕黄色，背景清晰。

WB注意事项：

样本处理：肠组织需用含1mM EDTA的RIPA裂解液（抑制钙依赖性蛋白酶），冰上裂解40分钟；

抗体稀释：一抗用5% BSA+TBST按1:1000稀释，4℃摇床孵育过夜，二抗（HRP标记山羊抗小鼠）1:5000室温孵育1小时。
