产品介绍Villin can cap, nucleate, sever and bundle actin in a calcium and phosphoinositide regulated manner. It is associated with the microvillar actin core bundle of intestinal and renal brush border implicated in adsorption. Villin is composed of six repeats, each containing 150 residues that together constitute the core domain followed by the carboxyl terminal headpiece domain of 87 residues. The core domain retains the calcium dependent capping nucleating and severing activity, whereas the headpiece domain contributes towards actin filament bundling and binding F actin, independently of Calcium.

Villin是一种肌动蛋白结合蛋白，属于gelsolin超家族，主要表达于肠上皮刷状缘、肾近曲小管、胆囊上皮等具有高度极性的细胞中。通过调节微丝组装与解聚参与细胞形态维持、物质转运和信号传导，在结直肠癌、胃癌、胆管癌等上皮源性肿瘤中表达异常，可作为肿瘤组织来源和分化程度的标志物。

实验操作关键要点

WB优化：

样本处理：肠组织样本建议使用含EDTA的RIPA裂解液（抑制钙依赖性蛋白酶），冰上裂解40分钟；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保92kDa条带与Marker清晰分离；

抗体孵育：一抗推荐1:800稀释（5% BSA+TBST），4℃摇床孵育过夜，二抗（HRP标记山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项：

抗原修复：采用EDTA缓冲液（pH9.0）高压修复3分钟（针对福尔马林固定石蜡切片），可增强胞膜与胞质信号；

内源性生物素阻断：若使用ABC检测系统，需用1% avidin封闭15分钟，再用1% biotin封闭15分钟（PBS洗涤间隔）；

阳性对照：建议使用正常小肠组织切片（刷状缘区域呈强阳性）。

常见问题与解决方案

IHC胞核染色干扰 固定时间过长（>24小时） 缩短甲醛固定时间至6-12小时，或使用4%多聚甲醛

WB无条带 抗原表位被封闭（如磷酸化修饰） 尝试去磷酸化处理（λ-磷酸酶37℃孵育30分钟）

IF信号弥散 透化过度（Triton X-100浓度>0.5%） 降低透化剂浓度至0.1-0.2%，缩短透化时间至5分钟
