产品介绍This gene encodes a member of the intermediate filament family. Intermediate filamentents, along with microtubules and actin microfilaments, make up the cytoskeleton. The protein encoded by this gene is responsible for maintaining cell shape, integrity of the cytoplasm, and stabilizing cytoskeletal interactions. It is also involved in the immune response, and controls the transport of low-density lipoprotein (LDL)-derived cholesterol from a lysosome to the site of esterification. It functions as an organizer of a number of critical proteins involved in attachment, migration, and cell signaling. Mutations in this gene causes a dominant, pulverulent cataract.[provided by RefSeq, Jun 2009]

WT1（Wilms Tumor 1）是一种锌指结构转录因子，在胚胎发育中调控肾脏、性腺等器官形成，成人组织中低表达或沉默。在肾母细胞瘤、急性髓系白血病、卵巢癌、乳腺癌等多种肿瘤中异常高表达，通过调控细胞周期（如抑制p53）、上皮-间质转化（EMT）等通路促进肿瘤发生发展，是临床诊断和预后评估的重要生物标志物。

实验操作关键要点

WB优化：

样本裂解：推荐使用含1% PMSF的RIPA裂解液，冰上裂解30分钟后12000g离心15分钟取上清；

转膜条件：目的蛋白52kDa，建议湿转300mA恒流60分钟（PVDF膜需甲醇活化）；

封闭与孵育：5%脱脂奶粉+TBST室温封闭1小时，一抗4℃孵育过夜（稀释比例根据样本丰度调整，低表达样本建议1:500起始）。

IHC-P注意事项：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波加热至95℃，持续15分钟后自然冷却至室温；

内源性过氧化物酶阻断：3% H₂O₂室温孵育10分钟，PBS洗涤3次×5分钟；

显色控制：DAB显色时间建议3-10分钟（镜下观察，避免背景过深）。

常见问题与解决方案

WB条带弥散或多带 抗体浓度过高/蛋白降解 降低抗体稀释比例（如1:2000）；样本添加蛋白酶抑制剂

IHC背景染色深 封闭不充分/抗体交叉反应 延长封闭时间至2小时；使用兔血清封闭（1:10稀释）

IF信号弱 透化不足/抗原修复不彻底 增加Triton X-100浓度至0.3%；延长修复时间至20分钟
