产品介绍This gene encodes an aspartic proteinase that belongs to the peptidase family A1. The encoded protein is a digestive enzyme that is produced in the stomach and constitutes a major component of the gastric mucosa. This protein is also secreted into the serum. This protein is synthesized as an inactive zymogen that includes a highly basic prosegment. This enzyme is converted into its active mature form at low pH by sequential cleavage of the prosegment that is carried out by the enzyme itself. Polymorphisms in this gene are associated with susceptibility to gastric cancers. Serum levels of this enzyme are used as a biomarker for certain gastric diseases including Helicobacter pylori related gastritis. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 1. [provided by RefSeq, Oct 2009]

靶点与功能

UCHL1/PGP9.5（Ubiquitin C-terminal Hydrolase L1）是泛素-蛋白酶体系统关键酶，主要功能包括：

神经元标志物：在中枢和外周神经元、神经内分泌细胞中高表达，占脑内可溶性蛋白的1-2%，是神经损伤修复和神经退行性疾病（如帕金森病、阿尔茨海默病） 的重要指标；

肿瘤诊断：在神经母细胞瘤、嗜铬细胞瘤、 Merkel细胞癌中特异性表达，阳性率＞85%，可用于与非神经源性肿瘤的鉴别；

泛素调控：通过水解泛素羧基末端酯键/肽键，参与蛋白降解和信号通路调控（如p53稳定性调节）。

实验操作关键要点

WB神经元样本检测：

样本处理：大鼠脑组织用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；推荐使用背根神经节（DRG）组织作为阳性对照；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.22μm PVDF膜（200mA恒流1小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光时间30秒（25kDa处可见清晰单一条带）。

IHC-P神经内分泌肿瘤染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），神经元胞浆信号显著增强；

阳性定位：正常脑组织神经元胞浆呈棕黄色阳性，神经母细胞瘤中可见弥漫性胞浆着色；

对照设置：必须包含脊髓组织（阳性对照）和肝细胞癌组织（阴性对照，排除非特异性结合）。

IF双标实验（神经再生研究）：

推荐与GFAP抗体（星形胶质细胞标志物）共染，使用Alexa Fluor 488（绿色，PGP9.5标记神经元）和594（红色，GFAP标记胶质细胞），激光共聚焦观察神经损伤后神经元存活状态。
