产品介绍The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin tightly associates with CDK9 kinase, and was found to be a major subunit of the transcription elongation factor p-TEFb. The kinase complex containing this cyclin and the elongation factor can interact with, and act as a cofactor of human immunodeficiency virus type 1 (HIV-1) Tat protein, and was shown to be both necessary and sufficient for full activation of viral transcription. This cyclin and its kinase partner were also found to be involved in the phosphorylation and regulation of the carboxy-terminal domain (CTD) of the largest RNA polymerase II subunit.

实验操作关键要点

WB检测通用流程：

样本处理：使用含磷酸酶抑制剂（PhosSTOP+Na₃VO₄） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清（避免磷酸化位点去磷酸化）；

抗体稀释：一抗用5% BSA+TBST按1:1000稀释（多克隆抗体建议预实验优化浓度），4℃孵育过夜；

内参选择：需同时检测总激酶蛋白（如总PLK1）及β-actin，确保磷酸化水平变化非因总蛋白表达差异。

IHC-P组织染色注意事项：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（PLK1/NEK9）或EDTA（pH9.0）高压修复（LKB1）；

阳性定位：

PLK1（Thr210）：肿瘤细胞核阳性（有丝分裂期）；

LKB1（Ser428）：肝细胞质/膜阳性；

NEK9（Thr210）：中心体及纺锤体区域阳性；

阴性对照：采用λ-磷酸酶处理的组织裂解液或敲除细胞系验证抗体特异性。
