Texas Red (Sulforhodamine 101) is an amphoteric rhodamineredfluorescent dye(excitation/emission: 586/605 nm). Texas Red is used extensively for investigating neuronal morphology and acts as a cell type-selective fluorescent marker of astrocytes both in vivo and in slice preparations^[1].

Texas Red (Sulforhodamine 101) does not label astrocytes in brainstem slices as strong and specific as in the hippocampus or cortex. To minimize excitatory side effects, the concentration of Texas Red has to be kept as low as possible or the labeling procedure can be performed after the actual experiment^[1].

In vivo, epileptic activity can be induced by intra-hippocampal injection of small volumes of 10 μM Texas Red or topical application of 100 μM^[1].

  以 1 mL 工作液为例，取 100 μL 20.8 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。

  将 0.9 g 氯化钠，完全溶解于 100 mL ddH2O 中，得到澄清透明的生理盐水溶液

  Description: Texas Red (Sulforhodamine 101) can be used as an astrocyte marker for hippocampal slices staining with red fluorescence.
  Method: For cell staining.
  1. Stain hippocampal slices with Texas Red (5 μM; 35 min; 34oC) in carbogenated Artificial CerebroSpinal Fluid (ACSF).
  2. Return hippocampal slices to dye-free ACSF for at least 30 min before imaging.
  3. Use a confocal laser scanning microscopy (Zeiss LSM 880) for image.
