Tumulosic acid 是一种三萜类物质，可抑制KLK5蛋白酶活性 (IC₅₀=14.84 μM)。Tumulosic acid 在体外抑制角质形成细胞中 LL-37 的蛋白水解过程 (≤10 μM)。

生物活性

Tumulosic acid, a triterpenoid, inhibitsKLK5 proteaseactivity (IC₅₀= 14.84 μM). Tumulosic acid suppresses the proteolytic processing of LL-37 in keratinocytes at ≤10 μM^[1].

体外研究
(In Vitro)

Tumulosic acid displays dose-dependent anti-KLK5 activity (IC₅₀=19.3 μM)^[1].
Tumulosic acid decreases in LL-37 production in human keratinocytes^[1].
Tumulosic acid shows IC₅₀values of 14.84 and 10.48 μM against KLK5 and trypsin proteases, while it exhibited a weak inhibitory effect on KLK7 and chymotrypsin C proteases (19.5% and 4.0%, respectively, at 100 μM)^[1].

Western Blot Analysis^[1]

Cell Line:	Normal human epidermal keratinocytes (NHEKs)
Concentration:	1 μM and 10 μM
Incubation Time:	23 h in the presence of 10 μM cycloheximide
Result:	Decreased LL-37 production in epidermal keratinocytes.
