N-(3,4,-Dimethoxycinnamoyl) anthranilic acid (3,4-DAA) is a synthetic derivative of the tryptophan metabolite anthranilic acid.
体外活性
N-(3,4,-Dimethoxycinnamoyl) anthranilic acid suppressed antigen-specific proliferation of MBP Ac1-11 TCR transgenic CD4+ T cells by arrested the cells in G1/S-phase. N-(3,4,-Dimethoxycinnamoyl) anthranilic acid (200 μM) decreased the release of IL-2, IFN-γ, and TNF-α and N-(3,4,-Dimethoxycinnamoyl) anthranilic acid (30 μM)increased the level of IL-4 and IL-10 in splenocytes from MBP Ac1-11 TCR transgenic T cells after antigen stimulation. N-(3,4,-Dimethoxycinnamoyl) anthranilic acid dose-dependently decreased IFNγ–induced cell surface expression of MHC class II and costimulatory molecules and suppressed the expression of inducible nitric oxide synthase and nitric oxide release from EOC20 cells and phosphorylation of STAT1α induced by IFNγ [1].
体内活性
N-(3,4,-Dimethoxycinnamoyl) anthranilic acid (300 mg/kg per day, p.o.) exhibited fewer and milder relapses and less severe disease compared to control animals in Mice with experimental autoimmune encephalomyelitis. In allograft immune rejection model, administration of N-(3,4,-Dimethoxycinnamoyl) anthranilic acid reduced histological severity of allograft immune rejection, decreased serum levels of TNF-α and IFN-γ, and raised serum levels of IL-10 [1][3].
