Collagenases are used at various concentrations for tissue dissociation, generally ranging from 0.1 to 5 mg/ml, and various time points depending on the desired affect. Several types of collagenases (Types 1, 2, 3, and 4) have been established based on the enzymatic profile. Collagenase Type I is a crude collagenase preparation that can be used for the isolation of primary cells or for tissue dissociation by enzymatic means. The preparation also may contain caseinase, clostripain, and tryptic activities. Type 1 is suggested for epithelial, liver, lung, and adrenal primary cell isolations. mCollagen fibers from sea cucumber showed partial depolymerisation into fibrils due to the fracture of proteoglycan interfibrillar bridges, as well as uncoiling of collagen fibrils after incubating in with 8 mL collagenase type I (71.25 U/mL) for 72 h.
