Mammalian membrane occupation and recognition nexus repeat-containing-2 (MORN2) is an orthologous protein. MORN2 is involved in the elimination of bacteria such as Legionella pneumophila, Staphylococcus aureus and Mycobacterium tuberculosis through the facilitation of LAP efficiency in macrophages. MORN2-mediated LAP requires ATG5 and Beclin-1 but not ULK1 and ATG13 to form single membrane-containing LAPosomes. MORN2-overexpressing macrophages enhance LAP efficiency (the ratio of LC3-positive phagosomes to total phagosomes) for these bacteria. Human MORN2 facilitates phagocytosis-mediated restriction of Mycobacterium tuberculosis, L. pneumophila, and S. aureus in macrophages. MORN2 promotes the recruitment of LC3, an autophagy protein also involved in phagocytosis, to M. tuberculosis-containing phagosomes and subsequent maturation to degradative phagolysosomes.
