Silvestrol is a naturally occuring flavagline analog isolated from the genus of Aglaia, it acts as a eukaryotic translation initiation factor 4A (eIF4A) inhibitor and exhibits anti-cancer activity in vitro and in vivo. Silvestrol induces early autophagy and apoptosis in human melanoma cells. Silvestrol exhibits direct anti-tumor activity while preserving innate and adaptive immunity against EBV-driven lymphoproliferative disease. Silvestrol exhibits significant in vivo and in vitro antileukemic activities and inhibits FLT3 and miR-155 expressions in acute myeloid leukemia.
