IDE1 (Inducer of definitive endoderm 1) is a novel, potent and cell-permeable inducer of definitive endoderm formation with EC50 of 125 nM). IDE1 induces Sox17 expression. It also activates TGF-β signaling and downstream Smad2 phosphorylation. IDE1 upregulates Nodal expression. It induces differentiation of mouse or human pluripotent stem cells (PSCs) by activating SMAD2 phosphorylation and NODAL expression (Borowiak et al.). At EC50 = 125 nM, SOX17 expression was induced in mouse ES cells.
