Letrozole是一种新型有效的(II)型芳香酶抑制剂,IC50值为11.5 nM。它属于可逆的非甾体类化合物,能抑制芳香化酶。没有证据表明它会影响肾上腺皮质类固醇[1]。
II型抑制剂的共同特征是具有可以结合到芳香酶血红素铁的杂环吡咯部分。其含有的1,2,4-三唑基部分可以协调细胞色素P450的血红素铁结构。同时,使用benzonitrile取代 letrozole可以模拟底物和雄烯二酮的独特的酶结构[2]。
Letrozole可以减少脊柱的突触和轴突生长,同时减少雌激素受体((ER()的表达。包括GAP-43在内的突触蛋白可以损害长时程增强[1]。Letrozole可以通过响应于雌激素(E)减少的反馈来促进下丘脑-垂体轴FSH的释放[3]。
