泛素特异性蛋白酶3(USP3)是uH2A和uH2B的去泛素化酶。体内USP3与染色质动态关联,去泛素化H2A / H2B。USP3的ZnF-UBP结构域介导uH2A-USP3相互作用。通过RNAi消除USP3的功能,导致S期延迟和DNA断裂的积累,随后激活DNA损伤检查点通路。响应于电离辐射,在g-H2AX DNA修复病灶中,uH2A重新分布和共定位(1),USP3对于uH2A/泛素结合物的去泛素化和g-H2AX去磷酸化具有重要作用(2)。USP3是H2A和H2B泛素化的新型调节剂,阐明了其在防止复制压力方面的作用,参与对DNA双链断裂的应答。
USP3已被表征为体外功能性的DUB,人DUB与酿酒酵母(S. cerevisiae)Ubp8最同源,参与调节H2B去泛素化4。
