Testosterone, the major androgen in males, acts by binding to the androgen receptor (AR), with an EC50 of 0.66 nM [1].
Testosterone is essential for the maintenance of spermatogenesis by binding to the AR, which is expressed in different cell types of the testis such as Sertoli cells and sperm cells [2]. Testosterone has also been suggested to play a role in mediating biological processes associated with bone metabolism [3].
In vitro incubation with testosterone (0, 2, 4, or 6 ng/mL) decreased the post-thaw acrosome integrity of ram (Ovis aries) sperm (from 68.1 ± 6.3% to 49.6 ± 3.9%) [2]. In RAW264.7 cells stimulated by receptor activator of nuclear factor kappa-Β ligand (RANKL), testosterone (1, 10 and 100 nM) decreased osteoclast formation in a dose-dependent manner, and, at 100 nM, significantly down-regulated the production of tumor necrosis factor-α (TNF-α) [3].
In a double-blind, placebo-controlled 6-week trial involving 74 patients, biweekly testosterone injections (200 mg as the initial dose, increased to 400 mg after 2 weeks), followed by 12 weeks of open-label maintenance treatment, were well tolerated and effective in the short-term treatment of symptoms of clinical hypogonadism in men with symptomatic human immunodeficiency virus illness, restoring libido and energy, alleviating depressed mood, and increasing muscle mass [4].
