  • Description: 5-TAMRA-SE (5-TAMRA-NHS ester) is a fluorescent agent with red fluorescence, it can be labeled to gene vectors/DNA for observing cellular uptake.
    Method: For cell labeling.
    1. The gene vectors/DNA NPs are labeled with 5-TAMRA-SE dye.
    2. Cells are seeded into the glass plates. After 20 h culture, the cell membrane is labeled with DIO dye and then treated with the same amount of the vectors/DNA NPs in 1 mL fresh RPMI 1640 medium without FBS.
    3. The living cells are immediately observed by TIRFM (Olympus, Japan). The fluorescence images are captured every 5 s, and the nanoparticles are analyzed by single-particle tracking (SPT) technique programmed by MATLAB software.
